Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells excluding mature erythrocytes and platelets. The cytoplasmic portion has tyrosine phosphatase activity, playing a key role in lymphocyte activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse bone marrow cells
Recommended use ≤ 0.4 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.

NovaFluor Compatibility Notice:

• Not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI)
• Recommended: Invitrogen LIVE/DEAD Fixable Dead Cell stains

Blocking Buffer Use:

• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining
• Add 5 µL per sample to antibody cocktails/master mixes before combining with cells
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). Absent on non-hematopoietic cells.
Functions:

• Regulates Src-family kinases in lymphocyte antigen signaling
• Essential in T/B-cell receptor signaling
• Modulates integrin-mediated signal transduction and apoptosis
• Influences cytokine receptor signaling via JAK kinase suppression
  Multiple isoforms arise from alternative splicing, differing in extracellular domains but sharing transmembrane and cytoplasmic regions.
