Product Specific Information

The 30-F11 monoclonal antibody reacts with all isoforms of mouse CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except mature erythrocytes and platelets. It plays a crucial role in lymphocyte activation via tyrosine phosphatase activity.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse bone marrow cells, ≤0.4 µg/test (100 µL final volume). Optimal cell number: 10⁵–10⁸ cells/test. Antibody titration recommended.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD45 (LCA) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It regulates Src-family kinases, T/B-cell antigen receptor signaling, and cytokine receptor pathways. CD45 isoforms arise from alternative splicing and differ by extracellular domain composition, influencing membrane localization and function. It plays roles in lymphocyte activation, survival, and apoptosis regulation.
