Mitochondrial glycerophosphate dehydrogenase (EC 1.1.99.5, GPD2)는 미토콘드리아 내막의 외측 표면에 위치하며, glycerol-3-phosphate (G-3-P)를 dihydroxyacetone phosphate (DHAP)로 전환하는 반응을 촉매합니다. 이 효소는 FAD를 환원시키며, 세포질 내 NAD-linked GPD (GPD1; MIM 138420)와 함께 glycerol phosphate shuttle을 형성하여, glycolysis 중 생성된 NADH의 환원력을 미토콘드리아로 전달하는 역할을 합니다.
