Target Information
The BBSome complex functions as a coat complex required for sorting of specific membrane proteins to the primary cilia. It is essential for ciliogenesis but dispensable for centriolar satellite function. This function is mediated by the Rab8 GDP/GTP exchange factor, which localizes to the basal body and interacts with the BBSome. Rab8(GTP) enters the primary cilium and promotes extension of the ciliary membrane. The BBSome associates with the ciliary membrane, binds to RAB3IP/Rabin8 (GEF for Rab8), and facilitates docking and fusion of carrier vesicles to the ciliary base. The BBSome complex, together with LTZL1, regulates SMO ciliary trafficking and contributes to sonic hedgehog (SHH) pathway regulation. It is required for BBSome ciliary localization but not for complex assembly.
