Target Information
Specialized cells at the midline, which separates the left and right halves of the CNS, have roles in directing growth cone behavior. In vertebrates and invertebrates, midline cells produce guidance cues such as nectins and slit, acting as attractants and repellents. Robo acts as a receptor for the repellent Slit and functions cell-autonomously. Non-crossing axons express high levels of Robo, while crossing axons show low expression before reaching the midline and high expression after crossing. Robo1 and Robo2 are human homologs of the Drosophila Roundabout protein, with Robo1 also homologous to the C. elegans gene sax3.
