Target Information
TMP21 is a ubiquitously expressed protein involved in vesicular targeting and protein transport. It is a component of the presenilin complex and modulates γ-secretase activity, but not ε-secretase cleavage of the amyloid precursor protein.
The presenilin complex includes APH1, nicastrin, PEN2, and presenilin-1, which together cleave amyloid precursor protein at γ- and ε-sites, leading to Abeta accumulation associated with Alzheimer’s disease.
Co-immunoprecipitation experiments indicate TMP21 participates in regulating this complex. Suppression of TMP21 via siRNA increases γ-secretase activity and Abeta production, showing TMP21’s modulatory role.
