Product Specific Information

The HIB19 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed on B cells throughout development except terminally differentiated plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, Leu13, and MHC class II, associating with the BCR. Signaling through CD19 induces tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (1.0 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which offers improved formulation over CellBlox. Add 5 µL per sample for optimal staining. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures enabling engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and on some myeloid leukemia cells. CD19 serves as a critical regulator of B cell activation, proliferation, and differentiation. It lowers the threshold for antigen receptor-dependent stimulation and can signal independently of BCR co-ligation. Mutations lead to hypogammaglobulinemia, while overexpression causes B cell hyperactivity.
