Product Specific Information
The 3D12 monoclonal antibody reacts with human CCR7 (also known as EBI-1 and CD197), a G-protein-coupled chemokine receptor with seven transmembrane domains. It functions as a receptor for 6Ckine/SLC (CCL19 and CCL21) and is involved in T cell and dendritic cell migration and maturation.
CCR7 expression distinguishes naive, central, and effector memory T cell populations and plays a role in trafficking memory T cells to inflamed tissues. During dendritic cell maturation, CCR7 expression increases, influencing chemotaxis, endocytosis, survival, and maturation. Ligand binding can downregulate surface expression.
Applications Reported: Flow cytometry and immunohistology (paraffin-embedded tissue).
Applications Tested: Flow cytometric analysis of human peripheral blood cells at ≤ 0.5 µg/test (100 µL final volume). Recommended to titrate for optimal performance.
Staining Condition: Incubate ≥ 45 minutes at 2–8 °C for optimal staining.
Purity: > 90% (SDS-PAGE)

Target Information
CCR7 is a member of the G protein-coupled receptor family and a chemokine receptor induced by Epstein-Barr virus (EBV). It is expressed in blood, bone marrow, lymph node, and intestine, particularly in lymphoid tissues and activated B/T lymphocytes. CCR7 regulates memory T cell migration to inflamed tissues and dendritic cell maturation.
Ligands include CCL19 (ECL) and CCL21. Related receptors include CCR1, CCR2A, CCR3, CCR4, CCR5, and the Duffy antigen.
