Target Information

C1orf106 is expressed in peripheral macrophages and intestinal myeloid-derived cells. It is essential for optimal PRR-induced signaling, cytokine secretion, and bacterial clearance. Upon stimulation of PRRs such as NOD2 or TLRs (TLR2, TLR3, TLR4, TLR5, TLR7, TLR9), it associates with YWHAQ/14-3-3T, leading to activation of MAP kinases and NF-κB signaling complexes that amplify immune responses.
In the intestine, it regulates adherens junction stability by controlling degradation of CYTH1 and CYTH2, likely acting as a substrate cofactor for SCF E3 ubiquitin-protein ligase complexes, thereby stabilizing junctions by limiting CYTH1-dependent ARF6 activation.
