Target Information

IL-33R, also known as ST2, is a receptor for IL-33 and a member of the IL-1 receptor family of innate receptors. Its extracellular domain consists of three Ig-like domains that interact with the co-receptor IL-1RAcP and the ligand IL-33. IL-33 binding activates the MyD88, IRAK1, IRAK4, and TRAF6 adaptor proteins, leading to NF-κB and MAPK pathway activation and subsequent release of cytokines such as IL-5, IL-6, IL-8, and IL-13.

Two common isoforms of IL-33R result from alternative splicing:

• ST2L: membrane-bound form
• ST2S: soluble form acting as a decoy receptor

IL-33R is primarily expressed in mast cells, basophils, and eosinophils. In mice, expression is also observed in ILC2 and Th2 cells. IL-33R signaling is implicated in various immune conditions including allergies, arthritis, atherosclerosis, and sepsis.
