Target Information
KIT (c-KIT)은 MGF(mast cell growth factor, stem cell factor)의 type 3 transmembrane receptor로 작용하는 proto-oncogene입니다. KIT은 feline sarcoma viral oncogene v-kit의 세포 동형체로 처음 확인되었습니다. KIT과 그 리간드는 다양한 조혈세포 및 비조혈세포의 성장과 활성화를 조절합니다.
KIT의 변이는 위장관 기질종양(GIST), 비만세포 질환, 급성 골수성 백혈병, piebaldism 등과 관련이 있습니다. 특히 D816V 활성 루프 변이에 의한 KIT 수용체의 탈조절은 비정상적인 비만세포 성장 억제를 위한 치료 전략의 초점이 되고 있습니다.
c-Kit은 조혈줄기세포, 생식세포, 비만세포 및 위장관의 Cajal 세포에서 발현됩니다. SCF(stem cell factor) 결합 시 c-Kit은 이합체를 형성하여 수용체를 활성화하고, 세포질 도메인 내 tyrosine 703을 포함한 다양한 타이로신 잔기를 자가인산화합니다. 이 과정은 Grb2 결합과 Ras/ERK 신호전달 경로의 활성화를 유도합니다. SCF/c-Kit은 PI3K, PLC-γ, JAK/STAT 등 다양한 하위 신호경로를 활성화할 수 있으며, c-Kit 수용체 활성화는 조혈, 줄기세포 유지 및 생식세포 형성에 필수적입니다.
