Product Specific Information

1 µg/mL of MA5-45250 was sufficient for detection of HIF1 alpha in 20 µg of CoCl₂-induced HeLa cell lysate by colorimetric immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.
Detects approximately 116 kDa. Specific for HIF1-alpha.

Target Information

HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under hypoxic conditions. HIF1 is a heterodimer composed of alpha and beta subunits, both belonging to the PAS family of transcription factors.
Under hypoxia, HIF1 activates transcription of over 40 genes involved in oxygen delivery and metabolic adaptation, including erythropoietin, glucose transporters, glycolytic enzymes, and VEGF.
HIF1-alpha also regulates hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. It interacts with p53 and MDM2 under hypoxic conditions, influencing protein degradation.
In humans, HIF1A is located on chromosome 14q and binds to p300/CBP to induce hypoxia-responsive genes such as VEGF.
Hypoxia contributes to the pathophysiology of diseases including ischemia, cancer, pulmonary hypertension, and chronic obstructive pulmonary disease.
