Target Information

RUVBL2 possesses single-stranded DNA-stimulated ATPase and ATP-dependent DNA helicase (5' to 3') activity. It is a component of the NuA4 histone acetyltransferase complex, which activates transcription by acetylating nucleosomal histones H4 and H2A. This modification influences nucleosome–DNA interactions and promotes interactions with proteins that regulate transcription positively.

The NuA4 complex contributes to transcriptional programs associated with oncogene-mediated growth induction, tumor suppressor-mediated growth arrest, replicative senescence, apoptosis, and DNA repair. RUVBL2, together with RUVBL1 and EP400, provides ATPase and helicase activities to the complex and plays essential roles in oncogenic transformation by MYC and transcriptional regulation via the LEF1/TCF1-CTNNB1 complex. It may also inhibit ATF2 transcriptional activity.
