Bruton tyrosine kinase (BTK) is a cytoplasmic tyrosine kinase belonging to the SRC-related TEC subfamily. Mutations in the BTK gene are linked to severe developmental blocks in human B-cell ontogeny and immunodeficiency disorders.
BTK interacts with toll-like receptor (TLR) family members such as TLR4, 6, 8, and 9, which are essential in innate and adaptive immunity. It also interacts with proteins involved in TLR4 signal transduction (MyD88, TIRAP, IRAK) but not TRAF-6, suggesting a role in lipopolysaccharide signal transduction.
