Target Information
RIPK1은 death receptor 결합, 병원체 인식 수용체(PRRs) 활성화, DNA 손상 등에 의해 염증 및 세포사멸 신호(프로그램된 괴사)를 전달하는 serine-threonine kinase입니다. TNFR1이 TNF-alpha에 의해 활성화되면 TRADD와 TRAF2가 수용체에 결합하고, RIPK1은 DAB2IP를 Ser-728에서 인산화하여 MAP3K5-JNK apoptotic cascade를 활성화합니다. TRAF2에 의한 Lys-63 연결형 유비퀴틴화는 MAPK 및 NF-kappa-B 경로의 하류 신호전달을 강화합니다. Polyubiquitinated RIPK1은 IKBKG/NEMO와 결합하여 NF-kappa-B 활성화를 유도하며, RHIM 도메인을 가진 다른 어댑터 단백질과 상호작용하여 유전자 활성화 및 세포사멸을 유도합니다. 특히 RIPK1과 RIPK3의 결합은 괴사 유도 복합체를 형성합니다.
