Target Information
TLR4는 선천 면역 활성화와 병원체 인식에 중요한 역할을 하는 톨유사수용체(TLR) 패밀리의 구성원입니다. 이 수용체는 태반 및 백혈구의 단핵구 아형에서 가장 풍부하게 발현되며, LPS에 의해 활성화됩니다. TLR4는 CD14, LY96과 함께 LPS 수용체 복합체를 형성하여 그람음성균의 병원체 관련 분자 패턴(PAMPs)을 인식하고, 면역 반응에 필요한 사이토카인 생성을 유도합니다. TLR4 유전자 돌연변이는 LPS 반응성 차이와 관련이 있으며, 여러 전사 변이가 존재합니다. TLR4는 말초혈액 단핵구 및 일부 B세포, 인간 태반에서 발현됩니다. TLR4 결핍 생쥐 연구에서는 주요 리간드가 LPS임을 확인하였으며, 그람음성균 패혈증에 대한 감수성이 증가함이 보고되었습니다.
