Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins linking TNF receptor superfamily members to signaling pathways, regulating cell death and stress responses. TRAF proteins share a conserved region enabling interactions with receptors and other TRAFs, activating diverse signal cascades.
TRAF2 and TRAF3 bind directly to CD40, a receptor involved in B cell immunity, and are essential for NF-kappa-B activation in mouse B lymphocytes. TRAF2, along with TRAF6, is required for CD40 signaling in non-hematopoietic cells and interacts with the lymphotoxin-beta receptor (LTβR) together with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
