Product Specific Information

Description:
The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. PD-1 is mainly expressed on activated T and B lymphocytes. Its ligands PD-L1 (B7-H1) and PD-L2 (B7-DC) are expressed on normal and activated antigen-presenting cells.

Costaining experiments indicate that eBioJ105 recognizes a different epitope than MIH4 (Product # 11-9969).

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry using human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test in 100 µL final volume.

Brilliant Violet™ 650 (BV650): Tandem dye emitting at 649 nm, for use with 405 nm violet laser-equipped cytometers. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.
Not Compatible: Methanol-based fixation.

Target Information

T lymphocytes initiate cell-mediated immune responses when stimulated by peptides bound to MHC molecules on antigen-presenting cells (APCs). PD-1 (PDCD-1) acts as an inhibitory receptor, binding to ligands PD-L1 and PD-L2 to downregulate immune activation. PD-1 deficiency leads to hyperactivated immune responses and autoimmune diseases. Despite its predicted molecular weight, PD-1 often migrates at higher molecular weight in SDS-PAGE.
