Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor is larger than 90 kDa and contains three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain.
AR acts as a ligand-activated transcription factor that binds active testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.

The AR signaling pathway is crucial for the development and function of male reproductive organs such as the prostate and epididymis, and also affects nonreproductive tissues including muscle, hair follicles, and brain.
AR is a phosphoprotein that regulates the MAP kinase pathway; inhibition of MEK1/2 correlates with changes in AR phosphorylation.
Abnormal AR signaling is associated with diseases such as prostate cancer, Kennedy’s disease, and male infertility. Mutations in this gene cause complete androgen insensitivity (CAIS).
