Specificity: This antibody is highly specific for murine IL-17A; in ELISA analysis, no cross-reactivity with IL-17B, IL-17C, IL-17D, IL-17E, or IL-17F was observed.
Target Information
Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine that promotes inflammatory responses in cell lines and is elevated in rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
IL-17A is a 32 kDa disulfide-linked homodimer consisting of 136 amino acids, belonging to a family of six related proteins (IL-17A–IL-17F) that signal through the IL-17 receptor (IL-17R/CDw217).
High levels of IL-17A homodimer are produced by activated peripheral blood CD4+ T-cells. IL-17A enhances expression of ICAM-1 in human fibroblasts and stimulates epithelial, endothelial, or fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2.
In the presence of IL-17A, fibroblasts can sustain proliferation of CD34+ hematopoietic progenitors and induce maturation into neutrophils. Mouse, rat, and human IL-17A can induce IL-6 secretion in mouse stromal cells, indicating cross-species receptor recognition.
IL-17A regulates NF-kappa B and MAP kinase activities, stimulates expression of IL-6 and COX-2, and enhances nitric oxide (NO) production.
