Product Specific Information

The 4B12 monoclonal antibody reacts with mouse CCR7 (CD197), a chemokine receptor for CCL19 and CCL21. CCR7 mediates lymphocyte migration into secondary lymphoid organs and plays a role in dendritic and B cell localization during immune responses.

Human CCR7 also marks central memory T cells (TCM), which express CCR7 and CD62L and reside in peripheral lymphoid organs. CCR7 contributes to thymocyte development, and its deficiency leads to disrupted thymic architecture and T cell development.

Staining Protocol

  1. Prepare cell suspension and block Fc receptors using purified anti-mouse CD16/32 (5 µg/million cells) for 15 min on ice.
  2. Without washing, add 4B12 antibody (1 µg/million cells) and incubate at 37°C or 2–8°C for 30 min.
  3. Wash cells once with 3 mL Flow Cytometry Staining Buffer and decant supernatant.
  4. Analyze by flow cytometry or proceed with secondary staining.

Note: Staining at 37°C yields brighter fluorescence and better resolution. Co-staining with CCL19-Fc may be hindered due to steric interference.

Target Information

CCR7 is a G protein-coupled chemokine receptor induced by Epstein-Barr virus (EBV). It is expressed in blood, bone marrow, lymph node, and intestine, particularly in activated B and T cells. CCR7 regulates migration of memory T cells and dendritic cell maturation. Its specific ligand is CCL19/ECL.
