Target Information
Focal Adhesion Kinase (FAK)는 125 kDa 비수용체 단백질 티로신 키나아제로, Src의 기질로 작용하며 인테그린 신호전달의 핵심 요소입니다. FAK는 세포 확산, 분화, 이동, 세포사 및 세포주기 G1에서 S기로의 전환에 중요한 역할을 합니다.
FAK는 중심 촉매 도메인과 C-말단 꼬리를 가지며, 이는 세포가 인테그린 수용체를 통해 세포외기질에 부착하는 국소 접착 부위(focal adhesion)에 위치하도록 합니다.
인테그린이 피브로넥틴과 결합하면 FAK의 티로신 인산화가 증가하며, Grb2 어댑터 단백질과 c-Src PTK가 FAK와 결합하고 ERK2 MAP kinase가 활성화됩니다.
또한 v-Src 변형 NIH3T3 세포에서는 FAK와 v-Src, Grb2, Sos의 결합이 세포 부착과 무관하게 일어납니다. Grb2 SH2 도메인은 인산화된 FAK의 Tyr925 부위에 직접 결합하며, 이는 변이 실험으로 확인되었습니다.
여러 전사 변이체가 존재하지만, 현재까지 완전 길이가 결정된 것은 세 가지입니다.
