Product Specific Information

The 37.51 monoclonal antibody reacts with the mouse CD28 molecule, a 45 kDa homodimer expressed by thymocytes, mature T cells, and NK cells.
CD28 is a ligand for CD80 (B7-1) and CD86 (B7-2) and acts as a potent costimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and increases cytotoxicity of CD3-activated T cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤ 0.5 µg/test.
A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL.
Cell number can vary (10⁵–10⁸ cells/test).
Careful titration recommended for optimal performance.

Note: Increased incubation times (30–45 min) may enhance staining.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T-cell surface glycoprotein of the immunoglobulin superfamily.
It provides the critical second activation signal to T cells by binding CD80 and CD86 on antigen-presenting cells.
CD28 prevents T cells from entering an anergic or apoptotic state.
In mice, CD28 is expressed on all CD4+ and CD8+ T cells in peripheral lymphoid organs and blood, and highly on immature thymocytes.
Diseases associated with CD28 dysfunction include mycosis fungoides and Sezary’s disease.
