Target Information
Members of the TNF superfamily regulate immune responses and induce apoptosis. Two novel members, BAFF (also known as BLyS, TALL-1, THANK, zTNF4) and APRIL (also known as TALL-2), have been identified.
- BAFF acts as a B-cell activator, inducing proliferation and immunoglobulin secretion.
- APRIL regulates both immunological and non-immunological cell growth.
Three receptors—BCMA, TACI, and BAFF-R—bind BAFF and APRIL.
TACI, similar to BCMA, plays a key role in regulating humoral immune responses, activating NF-κB and c-Jun N-terminal kinase pathways, and is implicated in B-cell–associated autoimmune diseases.
