Target Information
STAT3 (Signal Transducer and Activator of Transcription 3)는 STAT 단백질 계열에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동 후 전사 인자로 작용합니다. 이 단백질은 사이토카인 및 성장인자(Interferon-α, Interferon-γ, EGF, PDGF, IL6 등)에 반응하여 활성화됩니다.
JAK1 및 JAK2에 의한 티로신 705 잔기의 인산화는 STAT3의 이합체화, 핵 이동, DNA 결합 활성에 필수적입니다. 인간에서 STAT3 유전자는 염색체 17q에 위치하며, 다양한 세포 자극에 반응하여 유전자 발현을 매개하여 세포 성장 및 세포사멸 조절에 관여합니다.
STAT3는 Rac1 및 PIAS3와 상호작용하며, 세 가지 스플라이싱 변이체가 보고되어 있습니다. 마우스 모델에서 STAT3 유전자 결손은 배아 단계에서 치명적임이 확인되었습니다.
