Target Information

The cystatin superfamily is a well-established family of cysteine protease inhibitors.

• Type 1 (Intracellular): Cystatins A and B
• Type 2 (Extracellular): Cystatins C, D, E/M, F, G, S, SN, and SA
• Type 3 (Intravascular): Kininogens

All true cystatins inhibit cysteine peptidases of the papain family (e.g., cathepsins) and some also inhibit legumain family enzymes.
Cystatin SA, S, and SN are mainly found in saliva, and S and SN can also be expressed in tears, urine, and seminal fluid.
Cystatin C is expressed in brain, thymus, ovary, epididymis, and vas deferens.
Cystatin D protects against proteinases in the oral cavity, while Cystatin E/M and F moderate cathepsin inhibition.
Fetuins, part of the cystatin superfamily, are secretable proteins influencing osteogenesis, bone resorption, insulin and hepatocyte growth factor receptor regulation, and systemic inflammation response.
High and low molecular weight kininogens inhibit thrombin- and plasmin-induced thrombocyte aggregation.
