Product Specific Information
This CXNFT monoclonal antibody reacts to mouse NOS2 (inducible NOS, iNOS). Nitric oxide synthase enzymes catalyze the formation of nitric oxide from L-arginine through an NADPH- and oxygen-dependent mechanism.
Three isoforms of NOS are encoded by separate genes:
- NOS1 (neuronal NOS, nNOS)
- NOS3 (endothelial NOS, eNOS)
- NOS2 (inducible NOS, iNOS)
NOS2 is induced by bacterial endotoxins and inflammatory cytokines such as IFN-γ and TNF-α. It is expressed by myeloid-derived suppressor cells and M1 macrophages, but not M2 macrophages. NOS enzymes are active when forming homodimers, and NOS2 dimerizes at steady-state Ca²⁺ levels.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on mouse thioglycolate-elicited peritoneal exudate cells using Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
Recommended ≤0.125 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Titrate for optimal performance.
Fluorochrome Information:
Brilliant Violet™ 480 (BV480) emits at 479 nm and is intended for cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.
Note: BV480 is not compatible with methanol-based fixation.

Target Information
iNOS (Inducible Nitric Oxide Synthase) mediates NO production from L-arginine to L-citrulline.
- Function: Regulates vasorelaxation, neurotransmission, and cytotoxicity.
- Expression: Inducible in liver and various cells (macrophages, hepatocytes, smooth muscle cells).
- Induction: Stimulated by cytokines (IFN-γ, TNF, IL-1, IL-2) and LPS.
- Isoforms: NOS1 (neuronal), NOS3 (endothelial), NOS2 (inducible).
- Clinical relevance: iNOS dysfunction is associated with achalasia and impotence.
Three related iNOS pseudogenes are located within the Smith-Magenis syndrome region on chromosome 17.
