Product Specific Information

  • Immunohistochemistry (PFA fixed):
    Incubate antibody for 30 minutes at room temperature.
    Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Xeroderma pigmentosum type G (XPG) is a human genetic disease exhibiting extreme sensitivity to sunlight. The XPG protein, a member of the flap endonuclease 1 (FEN-1) structure-specific DNA repair endonuclease family, is an enzyme essential for DNA repair of solar ultraviolet (UV)-induced DNA damages. Human XPG nuclease makes the 3′ incision during nucleotide excision repair of DNA and cleaves model DNA bubble structures near the junction of unpaired DNA with duplex regions. A 29-amino acid region of human XPG (residues 981–1009) contains the PCNA binding activity. A conserved arginine (Arg992) is crucial for PCNA binding. Replication Protein A (RPA) binds specifically and directly to two excision repair proteins, XPA and XPG.
