Target Information
EIF2AK2 (PKR) is one of four kinases that phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to environmental stress. PKR plays a key role in antiviral defense via activation by double-stranded RNA produced during viral infection. It is involved in IFN-induced innate antiviral response, virus-induced apoptosis, and regulation of cell growth and differentiation. PKR mediates apoptosis through Fas expression and FADD/Caspase-8 signaling. It regulates multiple pathways including NF-kappaB, p53, p38, and PDGF signaling. PKR is implicated in tumor suppression, malignancy, and neurodegenerative diseases such as Alzheimer’s, Huntington’s, and ALS. Viruses can inhibit PKR activity to evade antiviral defense, such as influenza virus NS1 protein.
