Target Information
Caspase-activated DNase (CAD) cleaves chromosomal DNA during apoptosis. Apoptosis, a programmed cell death process that removes damaged or unnecessary cells during mammalian development, can be induced by various stimuli such as tumor necrosis factor family death factors, UV irradiation, anticancer drugs, and deprivation of survival factors.
During apoptosis, cells and nuclei undergo shrinkage and fragmentation, and chromosomal DNA is degraded into nucleosomal units. CAD, activated by caspases, exists as a complex with its inhibitor ICAD (Inhibitor of CAD). ICAD assists in proper folding of CAD and inhibits its DNase activity. Upon activation, caspases cleave ICAD, releasing CAD to enter the nucleus and cleave chromosomal DNA.
Activation of caspases is regulated by the balance between pro- and anti-apoptotic proteins of the Bcl-2 family, heat shock proteins, and inhibitors of apoptosis proteins (IAPs). Fourteen members of the caspase family have been identified, with seven mediating apoptosis.
