Product Specific Information
Description:
The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells, and binds glycosylated, fucosylated, sulfated sialylated glycoproteins including CD34, glycam-1, and MAdCam-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.125 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize titration for specific assays.
Fluorochrome Information:
Brilliant Violet™ 650 (BV650) emits at 649 nm, for use with a 405 nm violet laser. Ensure cytometer compatibility.
Staining Buffer Recommendation:
When using multiple polymer dyes (Super Bright, Brilliant Violet™, Brilliant UV™), use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.
Light Sensitivity:
Protect from light; this tandem dye is sensitive to photo-induced oxidation.
Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54).
Brilliant Violet™ 650 is not compatible with methanol-based fixation.

Target Information
Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It includes:
- C-type lectin binding domain
- EGF-like domain
- Two short consensus repeat (SCR) sequences
- Spacer, transmembrane, and cytoplasmic regions
CD62L is constitutively expressed on most leukocytes, including lymphocytes (except some memory T cells), monocytes, and polymorphonuclear cells.
