Description:
The monoclonal antibody TB28-2 recognizes the kappa light chain of human immunoglobulin. The Ig kappa chain (~22 kDa) consists of one variable and one constant region. Two kappa chains together with heavy chains form the complete immunoglobulin structure. Ig kappa expression is typically found on more than half of B lymphocytes.

Applications Reported:
This TB28-2 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume). Cell number may vary between 10⁵–10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Target Information

Immunoglobulin molecules consist of two heavy and two light chains (kappa or lambda, ~22.5 kDa). Kappa and lambda light chains have distinct constant regions, allowing differentiation by antigenic properties. The kappa to lambda ratio is approximately 70:30.
Antibodies to the kappa light chain are useful for identifying leukemias, plasmacytomas, and certain non-Hodgkin’s lymphomas. Demonstration of clonality in lymphoid infiltrates indicates malignancy.
