Description:
Monoclonal antibody P1H12 recognizes CD146 (also known as MUC18, s-endo, Endo-CAM, Mel-CAM), a member of the immunoglobulin superfamily. CD146 is expressed on endothelial cells, bone marrow fibroblasts, certain tumors (notably melanoma), mesenchymal stromal cells, and endometrial stromal cells.
It is also found on a subset of T cells but not on circulating endothelial cells (negative for CD31 and CD51/61). Expression can be induced on activated lymphocytes. The protein is heavily glycosylated, with over 50% of its mass from carbohydrates.

Cross-reactivity:
Reported to cross-react with mouse, rabbit, and canine, but not with rat.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested for normal human peripheral blood cells at 5 µL (0.125 µg) per test. A test is defined as the amount required to stain a sample in a final volume of 100 µL. Cell numbers may vary (10⁵–10⁸ cells/test).

Fluorophore Information:
eFluor® 450 is an alternative to Pacific Blue®, excited with a violet laser (405 nm) and emits at 445 nm. Ensure instrument compatibility.

Target Information

CD146 (MCAM):
An integral membrane glycoprotein of the immunoglobulin superfamily, expressed at cell–cell junctions of endothelial cells and other normal or malignant cell types.
Functions as a calcium-independent cell adhesion molecule and may also act in signal transduction, recruiting Fyn kinase and promoting tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.
CD146 contributes to endothelial cohesion, vascular integrity, and may facilitate melanoma metastasis via vascular interaction. It also functions as a receptor inducing FYN and PTK2 phosphorylation and transient intracellular calcium increase.
