Product Specific Information

The monoclonal antibody MPO455-8E6 recognizes myeloperoxidase (MPO), a protein within the azurophilic granules of myeloid cells. MPO is a multimeric enzyme composed of two 55 kDa and two 15 kDa subunits, associated with heme, giving a greenish color. It catalyzes the breakdown of hydrogen peroxide and oxidation of tyrosine, producing hypochlorous acid and tyrosyl radical, which contribute to neutrophil cytotoxic activity.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining of normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).

Recommended Protocol:

1. Stain whole blood with surface antibodies.
2. Lyse erythrocytes with 1X RBC lysis buffer (Product #00-4333-57).
3. Fix cells with IC Fixation Buffer (Product #00-8222-49).
4. Wash twice with Permeabilization Buffer (Product #00-8333-56).
5. Incubate with MPO455-8E6 for 30–60 minutes.
6. Wash and analyze by flow cytometry.

Use 5 µL (0.25 µg) per test. A test is defined as the amount of antibody for staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emitting at 445 nm. Ensure instrument compatibility.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation and is abundant in neutrophil azurophilic granules. It forms a tetramer of two light and two heavy chains and generates hypohalous acids crucial for neutrophil microbicidal activity.
Associated diseases include Myeloperoxidase Deficiency and Familial Alzheimer’s Disease type 1.
