Product Specific Information

The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 forms a complex with CD132 (common gamma chain) to create the IL-7 receptor (IL-7R). This type I glycoprotein (~75–80 kDa) is expressed by immature B cells, thymocytes at various stages, and most mature T cells, with transient down-regulation upon activation.

IL-7 binding triggers signal transduction via tyrosine kinase pathways including Jak/STAT. IL-7 is essential for lymphocyte development and homeostatic proliferation of peripheral T cells. IL-7R signaling also regulates TCR locus rearrangement in γδ T cells.

CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). CD4⁺CD25⁺ cells with low CD127 expression are enriched for Foxp3⁺ Tregs, exhibiting higher suppressive activity in vitro.

Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry on normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information

CD127 (Interleukin-7 Receptor alpha, IL7R) is a glycoprotein regulating lymphopoiesis. The active IL7R is an alpha/gamma chain heterodimer. The gamma(c) chain activates signal transduction, while the alpha chain mediates specific signaling events. CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4⁺/CD8⁺ T cells. Dysfunctions are associated with severe combined immunodeficiency (SCID).
