Target Information
VEGF (vascular endothelial growth factor)는 45 kDa의 호모다이머성, 이황화 결합된 당단백질로, 혈관신생(angiogenesis)에 관여하며 종양의 성장과 전이를 촉진합니다. VEGF는 혈관 내피세포의 생존, 분열, 화학주성, 투과성 증가 등 다양한 생리적 효과를 유도합니다.
VEGF 패밀리에는 VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, PIGF가 포함되며, 이들은 혈관신생과 관련된 세포 신호 조절의 핵심 역할을 합니다. 대부분의 종양은 혈관신생을 필요로 하며, VEGF는 이 과정의 주요 사이토카인으로 알려져 있습니다.
VEGF 유전자는 대체 스플라이싱을 통해 5가지 이상의 이소폼을 생성하며, 이들은 주로 내피세포에 발현되는 두 가지 티로신 키나아제 수용체(flt-1/VEGFR-1, flk-1/KDR/VEGFR-2)에 결합합니다. 또한, FLT1의 대체 스플라이싱 형태인 sFLT1은 VEGF를 높은 친화도로 결합하여 혈관신생을 억제하는 역할을 합니다.
VEGF의 과발현은 POEMS 증후군(Polyneuropathy, Organomegaly, Endocrinopathy, Monoclonal gammopathy, Skin changes, Crow-Fukase syndrome)과 관련이 있습니다.
