Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with pregnancy establishment and maintenance. PR exists as two forms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), resulting from alternative translation initiation sites. PR-B is the transcriptionally active form responsible for gene activation related to endometrial maintenance, pregnancy, and inhibition of ovulation. PR-A differs from PR-B by a 165 amino acid deletion at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity. In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes conformational change, dimerizes, and binds to progesterone response elements (PREs) to activate transcription. Null mutations in the PGR gene can cause reproductive abnormalities.
