Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with pregnancy. PR exists as a 94 kDa protein (Form A) or a 120 kDa protein (Form B) due to alternative translation initiation sites. PR-B is the transcriptionally active form, responsible for activating genes involved in endometrium maintenance, pregnancy, and inhibition of ovulation. PR-A differs from PR-B by a 165 amino acid deletion at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity. In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, conformational changes enable dimerization and binding to progesterone response elements (PRE), leading to transcriptional activation. Null mutations in the PGR gene result in reproductive abnormalities.
