LRP6은 Wnt/β-catenin 신호 전달 경로에서 필수적인 역할을 하며, Frizzled와 함께 코리셉터로 작용합니다. DKK1 및 DKK2와 높은 친화도로 결합하지만 DKK3과는 결합하지 않습니다. DKK1과의 상호작용은 Kremen 단백질에 의해 매개되는 LRP6의 엔도사이토시스를 통해 Wnt/β-catenin 신호를 차단합니다.
