Target Information

The NCF4 protein is a cytosolic regulatory component of the superoxide-producing phagocyte NADPH-oxidase complex, crucial for host defense mechanisms.
It is predominantly expressed in myeloid lineage cells and forms complexes with NCF2/p67-phox and NCF1/p47-phox, interacting with RAC1 to activate flavocytochrome b upon stimulation.
The PX domain binds phospholipid products of PI(3) kinase, implicating NCF4 in PI(3) kinase-mediated signaling. Phosphorylation of NCF4 negatively regulates enzyme activity.
Alternative splicing results in distinct isoforms.
