Target Information

Phosphofructokinase (PFK) is a major regulatory enzyme that catalyzes sugar metabolism, essential for energy production and normal cell function. In humans, three structural loci control PFK subunits—M (muscle), L (liver), and P (platelet)—which are variably expressed across tissues. Human fibroblasts and leukocytes express all three genes. PFK forms tetramers composed of combinations of these subunits, resulting in five possible isozymes (M4, M3L, M2L2, ML3, L4). It is an allosteric enzyme activated by ADP, AMP, or fructose bisphosphate, and inhibited by ATP or citrate. PFK catalyzes the key regulatory step in glycolysis. Deficiency may cause episodic illness with dark urine following strenuous exercise. Elevated red cell PFK levels are observed in individuals with Down syndrome.
