Target Information
p53 단백질은 세포 스트레스에 의해 활성화되는 서열 특이적 전사 인자입니다. DNA 손상이나 뉴클레오타이드 결핍 시 세포주기 정지 또는 세포사멸을 유도합니다. p53은 스트레스 신호에 반응하여 p21waf1 등 G1 또는 G2/M 정지에 관여하는 단백질 발현을 유도합니다.
구조적으로 p53은 N-말단 전사활성화 도메인, 중앙 DNA 결합 도메인, 올리고머화 도메인, C-말단 조절 도메인으로 구성됩니다. 여러 인산화 부위 중 Ser15 인산화는 p53 활성화 및 안정화에 중요합니다. Ser392 인산화는 인간 종양 형성과 연관되어 있습니다.
p53은 손상된 세포의 증식을 억제하여 항암 역할을 수행하며, 노화 및 산화 스트레스에도 관련이 있습니다. 정상 세포에서는 낮은 수준으로 존재하지만, 다양한 암 세포에서는 과발현되어 종양 형성에 기여합니다. TP53 유전자의 변이는 여러 암 및 Li-Fraumeni 증후군과 같은 유전성 암 질환에서도 발견됩니다.
