Product Specific Information
Store vial at -20°C prior to opening. Aliquot contents and freeze at -20°C or below for extended storage. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.
This antibody recognizes the Me1, Me2, and Me3 forms of Lys27 in human Histone H3. BLAST analysis indicates cross-reactivity with Human, Mouse, Rat, and C. elegans based on 100% sequence homology. Cross-reactivity with Histone H3 [methyl Lys27] from other sources has not been determined.
Target Information
The nucleosome consists of 146 bp of DNA wrapped around an octamer of histone proteins (H2A, H2B, H3, and H4). Histone proteins are covalently modified at specific residues, mainly in the N-terminal tail, through processes such as acetylation, methylation, SUMOylation, ubiquitinylation, arginine methylation, citrullination, serine/threonine phosphorylation, and proline isomerization.
Lysine residues can be mono-, di-, or tri-methylated, and arginine residues can be mono- or dimethylated (symmetrically or asymmetrically). These modifications are temporally and cell-specifically regulated, influencing gene activation or repression.
- H3K4Me3 is associated with transcriptional activation at promoter regions.
- H3K4Me1 is found at enhancer sequences.
- H3K9Me1, H3K27Me1, and H4K20Me1 are linked to gene activation.
- H3K9Me2/Me3, H3K27Me2/Me3, and H4K20Me2/Me3 are linked to gene repression.
Monomethylation of H4K20 by SET8 is essential for genome replication and stability. Loss of this mark leads to DNA breaks and p53 activation, preventing abnormal mitosis. In mammalian stem cells, Xist expression inhibits H4K20me1 formation, demonstrating a link between chromatin regulation and stem cell differentiation.
