Product Specific Information

Description:
The 104 monoclonal antibody reacts with mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.2-expressing mouse strains such as C57BL/6, BALB/c, C58, DBA/1, DBA/2, C3H/He, CBA, 129, A, and AKR. CD45.2 is expressed by all leukocytes in these strains.

Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes.
Recommended usage ≤0.5 µg/test in 100 µL final volume.
Cell count may range from 10⁵ to 10⁸ cells/test.
Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample, regardless of the number of NovaFluor dyes.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing novel nucleic acid dye structures for engineered fluorescent signatures with minimized spillover and spread.

Target Information

CD45.2 (LCA, leukocyte common antigen)
A receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins).
Plays a crucial role in lymphocyte development, antigen signaling, and regulation of Src-family kinases.
Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
CD45.2 also promotes cell survival and participates in integrin-mediated signaling and apoptosis-related DNA fragmentation.
CD45RA is one isoform with restricted expression among lymphoid cell subtypes.
