Target Information

SHANK proteins are major scaffold components of the postsynaptic density, interacting with multiple proteins to orchestrate dendritic spine and synapse formation, maturation, and maintenance. They interconnect NMDA-type and metabotropic glutamate receptors through complexes with GKAP/PSD-95 and HOMER, linking to the actin cytoskeleton. SHANK3 regulates actin dynamics, neuronal growth cone motility, and receptor-mediated synaptic transmission and plasticity. It may be required at early synapse formation stages and modulated by IGF1 for maturation.
