Target Information

The tumor necrosis factor (TNF) receptor superfamily consists of several type I integral membrane glycoproteins sharing homology in their cysteine-rich extracellular domains. Members include TNF-RI, TNF-RII, and CD40. Ligands for these receptors can be small secreted proteins (e.g., TNF) or type II integral membrane proteins (e.g., CD40L). Activation of TNF-R or CD40 induces nuclear translocation of NFκB, a process involving members of the TRAF (TNF receptor-associated factor) family. Four TRAF members—TRAF1, TRAF2, TRAF3, and TRAF4—have been identified. TRAF4, originally termed CART1, is specifically expressed in breast carcinomas and localized to the nucleus in such tissues.
