Target Information
The tumor necrosis factor (TNF) receptor superfamily is composed of several type I integral membrane glycoproteins that share homology in their cysteine-rich extracellular domains. Members include TNF-RI, TNF-RII, and CD40. Ligands for these receptors may be small secreted proteins such as TNF, or type II integral membrane proteins like CD40L. Although the signal transduction mechanism of the TNF receptor superfamily is not fully understood, activation of TNF-R or CD40 induces nuclear translocation of NFκB. Members of the TRAF (TNF receptor-associated factor) family, including TRAF1, TRAF2, TRAF3, and TRAF4, are involved in this process. TRAF4, originally termed CART1, is specifically expressed in breast carcinomas and localized to the nucleus in these tissues.
