Target Information

HIF1-alpha (HIF1A) is a subunit of HIF1, a transcription factor found in mammalian cells under reduced oxygen tension. HIF-1 is a heterodimer consisting of alpha and beta subunits, both belonging to the basic-helix-loop-helix Per-aryl hydrocarbon receptor nuclear translocator-Sim (PAS) family. It regulates the adaptive response to hypoxia by activating transcription of over 40 genes, including erythropoietin, glucose transporters, glycolytic enzymes, VEGF, and others involved in oxygen delivery and metabolic adaptation.
HIF1-alpha also regulates hypoxia-mediated apoptosis, cell proliferation, and tumor angiogenesis. Under hypoxia, it interacts with p53 and undergoes MDM2-mediated ubiquitination and degradation. It also induces NOX4 to maintain ROS levels and support hypoxia-induced proliferation.
In humans, HIF1A is located on chromosome 14q. The C-terminal of HIF1A binds to p300, forming p300/CBP-HIF complexes that induce hypoxia-responsive genes, including VEGF.
Hypoxia plays a critical role in diseases such as myocardial and cerebral ischemia, cancer, pulmonary hypertension, congenital heart disease, and COPD.
