Target Information

Neurofilaments (NFs) are a type of intermediate filament (IF) expressed almost exclusively in neuronal cells, particularly in large axons. NFs in vertebrates consist of three polypeptide chains: neurofilament medium protein (NF-M), high (NF-H), and light protein (NF-L). These share a coiled-coil core domain but differ in terminal sequences, especially in the C-termini, which form flexible extensions linking NFs to each other and other cytoplasmic elements.
NF-M tail-mediated interactions are critical for axonal size and cytoskeletal architecture, influenced by phosphorylation and O-GlcNAcylation. Antibodies against NF subunits serve as useful neuronal markers due to their abundance, neuron-specific expression, and biochemical stability.
