Target Information
Cyclin D1 (PRAD-1, bcl-1)는 세포 주기 조절의 핵심 단백질로, CDK4 및/또는 CDK6과 결합하여 Rb 단백질을 인산화함으로써 세포 주기 G1/S 전이를 조절합니다. Cyclin D1은 다양한 인간 종양에서 과발현되는 원암유전자(proto-oncogene)로 알려져 있으며, 특히 mantle cell lymphoma(MCL)에서 특징적으로 발현됩니다.
Cyclin D1은 세포질 및 핵에 존재하며, 성장 인자 자극에 의해 합성되어 CDK4 또는 CDK6과 복합체를 형성합니다. Cyclin D1-CDK 복합체는 Rb 단백질의 억제 기능을 비활성화시키고, p21Cip1 및 p27Kip1과 같은 CDK 억제 단백질을 조절합니다. 성장 인자 자극이 없을 경우 Cyclin D1은 불안정하며, 인산화에 의해 유비퀴틴 매개 분해가 유도되어 G1/S기 정지에 관여합니다.
